ReHMGB1, a blood-borne driver of systemic aging revealed: HMGB1 never ages!

A team of researchers led by South Korean scientists has demonstrated that ReHMGB1-fully reduced HMGB1, is not only a multifunctional mediator of inflammation and tissue repair but also a potent inducer of aging-related cellular phenotypes.
ReHMGB1 can travel through the bloodstream to distant tissues, where it triggers cellular senescence.

Fully reduced HMGB1 emerges as a pro-geronic factor in aging
This finding is particularly striking because it establishes extracellular fully reduced HMGB1, as a redox-dependent pro-geronic factor, capable of converting local senescent signals into systemic aging.

In contrast, the oxidized disulfide form of HMGB1 did not produce the same effects, underscoring the unique biology of the fully reduced redox state. This is the first evidence that ReHMGB1 alone drives systemic senescence propagation.
Perhaps most importantly, the study offers hope for therapeutic intervention: blocking extracellular HMGB1 with neutralizing antibodies effectively suppressed senescence, reduced systemic inflammation, and promoted tissue regeneration in a muscle injury model in middle-aged mice. This suggests that HMGB1-targeting strategies could represent a promising avenue for mitigating age-related pathologies.
